Coiled Therapeutics plc (AIM: COIL) (OTCQB: COTXF), a clinical-stage precision oncology company, announced on Monday that it has dosed the first patient in the US with an optimised lipid-based oral formulation of its Phase I cancer therapy AO-252, with initial post-dose monitoring showing no treatment-related adverse safety findings.
The new softgel formulation is designed to improve the consistency and dose-proportional absorption of AO-252, a first-in-class brain-penetrant TACC3 inhibitor being evaluated in patients with advanced solid tumours.
This formulation follows encouraging Phase I results announced on 7 July 2026, in which twice-daily dosing with the tablet formulation achieved an 80% clinical benefit rate in a cohort of five heavily pre-treated patients, including tumour reductions in ovarian and endometrial cancer.
Coiled Therapeutics expects initial safety and pharmacokinetic data from the new formulation in the fourth quarter of 2026, with positive results expected to support planned Phase I/II dose-expansion cohorts in ovarian and prostate cancer and ongoing discussions with potential pharmaceutical partners.
